He was baptized on December 20, 1584, probably in Etton, Eastriding, Yorkshire, England and died on November 11, 1653 in Barnstable, Plymouth Colony, Massachussets.
He was the son Thomas Lothrop and Mary Howell.
Reverend John Lothropp was admitted to the degree Bachelor of Arts at Queen´s College in 1606 and to the Master of Arts in 1609; was pastor of the First Independent or Congregational Church at London; was in jail for non-conformist preaching and was sent into exile; arrived with his family on the ship Griffin at Boston September 18, 1634 with about 100 passengers and livestock and proceeded to Scituate, Plymouth Colony; removed to Barnstable in 1639 and was first minister there.
First he married Hannah Howse (House), born in Eastwell, Kent, England and died on February 16, 1633 in London, England, probably the daughter of Reverend John Howse and Alice Lloyd, on October 10, 1610 in Eastwell, Kent, England.
Second he married Ann, born on July 16, 1616 in Lavenham, Suffolk, England and died on February 25, 1687-88 in Barnstable, Barnstable, Dominion of New England (from 1686 to 1689), probably on September 27, 1634 in Scituate, Plymouth Colony (from 1620 to 1691).
John and Hannah Howse had the following children:
- Thomas was baptized on February 21, 1612-13 in Eastwell, Kent, England and died in 1707.
- He married Sarah Learned (Ewer), daughter of William Learned and Goodith Hall, born before January 18, 1607 in Bermondsey, Surry, England and died in 1652 in Barnstable, Plymouth Colony on December 11, 1639 in Boston, Suffolk, Massachusetts.

- Jane was baptized on September, 29 1614 in Egerton, Kent, England and died on October 26, 1678 in Barnstable, Barnstable, Plymouth Colony.
- She married Samual Fuller, son of Edward Fuller (Mayflower) and Ann, born about 1609 and died on October 31, 1683 in Barnstable, Barnstable, Massachusetts Bay Colony, on April 8, 1638 in Scituate, Plymouth Colony.

- Barbara was baptized on October 31, 1619 in Egerton, Kent, England and died probably 1638.
- She married John Emerson, son of John Emerson and Elizabeth Brewster on July 19, 1638 in Duxbury, Massachusetts.

- Elizabeth Scrudder, born before 1625 and died about 1688, daughter of John Scrudder and Elizabeth Stoughton, on November 28, 1644 in Barnstable, Barnstable, Plymouth Colony.
